Effects of auxin and cytokinin on production of diosgenin by callus cultures of Trigonella foenum graecum L
Abstract
Trigonella foenum-graecum L. (fenugreek) is a foremost medicinal plant known for its steroidal saponin diosgenin, a forerunner for several pharmaceutical steroids. The present study aimed to elevate diosgenin production through in vitro callus culture by employing plant growth regulators. Leaf explants cultures were raised from seedlings that had sprouted in vitro on Murashige and Skoog (MS) medium that had varied amounts of benzylaminopurine (BAP), 2,4-dichlorophenoxyacetic acid (2,4-D), and their combinations. Over a six-week culture period, we looked at callus induction, growth response, and diosgenin development.
Diosgenin content in callus culture tissues was quantified and analyzed using HPTLC (High Performance Thin Layer Chromatography). Among individual hormones, MS medium containing 0.5 mg/L BAP resulted in maximum diosgenin accumulation (1.593 mg/g fresh weight of the callus). The maximum diosgenin production i.e. (1.680 mg/g fresh weight of the callus culture) was recorded in the cultures grown on MS medium supplemented with a combination of BAP and 2,4-D at 0.5:1.0 mg/l. The study exemplify that appropriate hormonal manipulation outstandingly increases diosgenin production in fenugreek callus cultures, specifying the potential of in vitro culture systems as a most authentic alternative for large-scale production of this medicinally important secondary metabolite
