Abstract

This study was aimed to develop a protocol of callus initiation and propagation of four Rare, Endangered and Threatened (RET) species of arid and semi-arid regions namely, Capparis decidua, Commiphora wightii, Leptadenia reticulata and Ziziphus nummularia. Parameters considered for study: explant type, effects of sterilization method, media composition and plant growth hormones. Murashige and Skoog’s Media supplemented with 3% Sucrose and 0.8% Agar was used to initiate and proliferate callus in C. decidua, L. reticulata and Z. nummularia whereas Gamborg’s B5 media along with 3% Sucrose and 0.8% Agar was used for callus initiation as well as proliferation of C. wightii. The callus obtained from all the above species was kept on proliferation media for different periods without sub-culturing: 4 weeks, 8 weeks and 12 weeks to observe the viability of callus in media-depriving conditions and to further study their storage period as a viable strategy for medium-term conservation.
